The advance builds on work published in 2015 when researchers at Duke University grew the first functioning human muscle tissue from cells obtained from muscle biopsies. The ability to start from cellular scratch using non-muscle tissue will allow scientists to grow far more muscle cells, provide an easier path to genome editing and cellular therapies, and develop individually tailored models of rare muscle diseases for drug discovery and basic biology studies. The results appear in the journal Nature Communications.\n\n“Starting with pluripotent stem cells that are not muscle cells, but can become all existing cells in our body, allows us to grow an unlimited number of myogenic progenitor cells,” said Nenad Bursac, professor of biomedical engineering at Duke University. “These progenitor cells resemble adult muscle stem cells called ‘satellite cells’ that can theoretically grow an entire muscle starting from a single cell.”\n\nIn their previous work, Bursac and his team started with small samples of human cells obtained from muscle biopsies, called “myoblasts,” that had already progressed beyond the stem cell stage but hadn’t yet become mature muscle fibers. They grew these myoblasts by many folds and then put them into a supportive 3-D scaffolding filled with a nourishing gel that allowed them to form aligned and functioning human muscle fibers.\n\nIn the new study, the researchers instead started with human induced pluripotent stem cells. These are cells taken from adult non-muscle tissues, such as skin or blood, and reprogrammed to revert to a primordial state. The pluripotent stem cells are then grown while being flooded with a molecule called Pax7 — which signals the cells to start becoming muscle.\n\nAs the cells proliferated they became very similar to –but not quite as robust as — adult muscle stem cells. While previous studies had accomplished this feat, nobody has been able to then grow these intermediate cells into functioning skeletal muscle.\n\nThe Duke researchers succeeded where previous attempts had failed.\nhttps://dailyaccord.com/wp-content/uploads/2018/01/engineersgro.mp4?_=1 \n\n“It’s taken years of trial and error, making educated guesses and taking baby steps to finally produce functioning human muscle from pluripotent stem cells,” said Lingjun Rao, a postdoctoral researcher in Bursac’s laboratory and first author of the study. “What made the difference are our unique cell culture conditions and 3-D matrix, which allowed cells to grow and develop much faster and longer than the 2-D culture approaches that are more typically used.”\n\nOnce the cells were well on their way to becoming muscle, Bursac and Rao stopped providing the Pax7 signaling molecule and started giving the cells the support and nourishment they needed to fully mature.\n\nIn the study, the researchers show that after two to four weeks of 3-D culture, the resulting muscle cells form muscle fibers that contract and react to external stimuli such as electrical pulses and biochemical signals mimicking neuronal inputs just like native muscle tissue. They also implanted the newly grown muscle fibers into adult mice and showed that they survive and function for at least three weeks while progressively integrating into the native tissue through vascularization.\n\nThe resulting muscle, however, is not as strong as native muscle tissue, and also falls short of the muscle grown in the previous study that started from muscle biopsies. Despite this caveat, the researchers say this muscle still holds potential that the stronger, older relative does not.\n\nThe pluripotent stem cell-derived muscle fibers develop reservoirs of “satellite-like cells” that are necessary for normal adult muscles to repair damage, while the muscle from the previous study had much fewer of these cells. The stem cell method is also capable of growing many more cells from a smaller starting batch than the biopsy method.\n\nBoth of the advantages point toward a possibility of using this new method for regenerative therapies and for creating models of rare diseases for future studies and individualized health care.\n\n“The prospect of studying rare diseases is especially exciting for us,” said Bursac. “When a child’s muscles are already withering away from something like Duchenne muscular dystrophy, it would not be ethical to take muscle samples from them and do further damage. But with this technique, we can just take a small sample of non-muscle tissue, like skin or blood, revert the obtained cells to a pluripotent state, and eventually grow an endless amount of functioning muscle fibers to test.”\n\nThe technique also holds promise for being combined with genetic therapies. Researchers could, in theory, fix genetic malfunctions in the induced pluripotent stem cells derived from a patient and then grow small patches of completely healthy muscle. While this could not heal or replace an entire body’s worth of diseased muscle, it could be used in tandem with more widely targeted genetic therapies or to heal more localized problems.\n\nThe researchers are now refining their technique to grow more robust muscles and beginning work to develop new models of rare muscle diseases.",

      "body": "![](https://steemitimages.com/DQmfJQukLEJ1csqdJ6hsezhrBGW7UAzYpXGgQuqNdCpcd4J/%D0%B8%D0%B7%D0%BE%D0%B1%D1%80%D0%B0%D0%B6%D0%B5%D0%BD%D0%B8%D0%B5.png) \n\n\nWow.\nDAVID NIELD\n6 JAN 2018\n\nWe're used to dealing with three physical dimensions and one extra dimension of time as we move through the Universe, but two teams of scientists have shown that a fourth spatial dimension could reach beyond the limits of up and down, left and right, and forwards and backwards.\n\nAs you might expect given this is bending the laws of physics, the experiments involved are partly theoretical and very complex, and touch on our old friend quantum mechanics.\n\nBy placing together two specially designed 2D setups, two separate teams of researchers - one in Europe and one in the US - were able to catch a glimpse of this fourth spatial dimension through what's known as the quantum Hall effect, a certain way of restricting and measuring electrons. \n\n\"Physically, we don't have a 4D spatial system, but we can access 4D quantum Hall physics using this lower-dimensional system because the higher-dimensional system is coded in the complexity of the structure,\" a researcher with the US-based team, Mikael Rechtsman from Penn State University, told Ryan F. Mandelbaum at Gizmodo.\n\n\"Maybe we can come up with new physics in the higher dimension and then design devices that take advantage the higher-dimensional physics in lower dimensions.\"\n\nIn other words, just as a 3D object casts a 2D shadow, scientists have managed to observe a 3D shadow potentially cast by a 4D object – even if we can't actually see the 4D object itself. That could unlock some new findings in the very fundamentals of science.\n\nThanks to some rather advanced calculations – which won the Nobel Prize for Physics in 2016 – we know that the quantum Hall effect points to the existence of a fourth spatial dimension.\n\nWhat these new experiments do is give us a picture of the effects that this fourth dimension might have.\n\nThe European team's setup involved atoms cooled down close to absolute zero and placed in a 2D lattice through the user of lasers, described as \"an egg-carton-like crystal of light\" by the researchers.\n\nWith the addition of extra lasers, the team was able to implement a quantum \"charge pump\" to excite the trapped atoms and get them moving. Slight variations in the movement spotted by the researchers match up with how a 4D quantum Hall effect would ripple out – adding weight to the possibility that a fourth spatial dimension can be somehow accessed.\n\nThe US experiment also used lasers, this time to control light as it flowed through a block of glass. By manipulating the light to simulate the effect of an electric field on charged particles, again the consequences of a 4D quantum Hall effect could be observed.\n\nOf course, we can't physically access this 4D world – we're stuck in 3D space – but scientists think quantum mechanics could somehow give us a picture of it, thus enhancing our limited understanding of the Universe.\n\nAnother way to think about it, courtesy of this video, is if we were video game characters from a 2D platform, and suddenly wandering around in a 3D game.       "body": "That's 2 questions to be honest:\n- Yes, BTC can be dethroned, \"all\" it takes is one of the other coins to explode into a much higher marketcap. With the recent dynamics since start of 2018, that is entirely possible within the next 12 months ;-)\n- As for the price of BTC, I think a lot of that depends on how quickly things like SegWit and LN get active and used by most of the network. As long as that is not the case, the price will either slowly climb or remain as is. When it does get implemented and transaction costs/speeds improve, BTC price will be positively impacted.",
